VLA15, developed by Valneva and Pfizer, is a Lyme disease vaccine candidate targeting Borrelia burgdorferi. A previous Lyme vaccine, LYMErix by SmithKline Beecham, was licensed in 1998 but withdrawn in 2002 due to low sales and concerns about autoimmune side effects that were never confirmed. The withdrawal discouraged Lyme vaccine development for years. VLA15 targets the outer surface protein A OspA, the same target as LYMErix, but is designed to avoid the theoretical autoimmune risk. Phase 3 trials began in 2022. Lyme disease is the most common vector borne disease in the US, with an estimated 476,000 people diagnosed and treated annually. The vaccine would be welcomed in endemic areas of the Northeast, upper Midwest, and parts of Europe.
