Group A Streptococcus vaccines target Strep A, which causes pharyngitis, skin infections, rheumatic fever, and rheumatic heart disease. Rheumatic heart disease affects approximately 40 million people and kills approximately 300,000 annually, mostly in developing countries. Strep A vaccine development has been challenging. Concerns about vaccine induced rheumatic fever, which led to the discontinuation of early vaccine candidates in the 1960s, slowed development. The Strep A Vaccine Global Consortium SAVAC aims to accelerate Strep A vaccine development. Multiple candidates are in clinical trials, including conjugate vaccines, protein based vaccines, and mRNA vaccines. The Australian government has invested in Strep A vaccine research, motivated by the high burden of rheumatic heart disease in Indigenous communities.
