Uropathogenic E coli vaccine development targets the leading cause of urinary tract infections UTIs. UTIs affect approximately 150 million people annually. Recurrent UTIs are a significant problem for women, requiring repeated antibiotic courses that contribute to resistance. Several vaccine candidates have been developed including uromune, a sublingual vaccine containing killed E coli and other uropathogens, and ExPEC4V, a conjugate vaccine targeting E coli extraintestinal pathogenic strains. Clinical trials have shown mixed results. A vaccine preventing recurrent UTIs would significantly reduce antibiotic use and improve quality of life. The challenge is that UTIs are caused by multiple bacterial species, and a broad spectrum vaccine may be necessary.
