The Matrix M adjuvant used in the Novavax COVID vaccine is derived from the bark of the Quillaja saponaria tree, an evergreen native to Chile. The tree, also called soap bark tree, has been used medicinally for centuries. Saponins from the bark activate innate immune cells through multiple pathways. Matrix M forms nanoparticles that enhance antigen uptake by immune cells. The Chilean government regulates Quillaja harvest to ensure sustainability. Matrix M is also used in the R21 malaria vaccine and has been tested with influenza vaccines. The use of a natural product from a Chilean tree in a COVID vaccine developed by an American company illustrates the global nature of vaccine development. The Matrix M technology was also licensed to the Serum Institute of India for Covovax production.
